Background: Cryopreservation refers to the storage of a living organism at ultra-low-temperature for long-term preservation of plant germplasm. The effect of cryopreservation on the efficiency of exogenous gene genetic transformation and expression level were studied herein. In this work, transgenic Arabidopsis thaliana were successfully conserved in vitro by cryopreservation methods. Germination of Ipomoea cynanchifolia seeds after storage at different temperatures and scarification. Abstract ? Ipomoea cynanchifolia Meisn., Convolvulaceae, is a wild relative of Ipomoea batatas (L.) Lam. (sweet potato). In this study, seeds with 7.8% water content were stored in a germinator (25 °C), a freezer (-18 °C) and in liquid nitrogen (-196 °C) for seven days. Cryopreservation is widely used for long-term conservation of various tissues, embryos or gametes. However, few studies have described cryopreservation of invertebrate primary cell cultures and more particularly of marine invertebrate somatic cells. Due to stock declines there is a growing interest in farming of European hake Merluccius merluccius. Thus, knowledge of its sperm biology is of importance not only for purposes of broodstock management, but also for the development of sperm preservation techniques. Hake sperm were collected from mature males caught during the summer-early autumn waters off western Norway and during the winter-early spring in the Bay of Biscay (France). Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique. Cryopreservation is a valuable tool for genetic improvement programs. Several bivalve mollusc species have already been the subject of such programs and the Tahitian black pearl oyster industry is now planning the development of selective breeding for desirable traits in Pinctada margaritifera. The ability to cryopreserve spermatozoa would, therefore, offer significant benefits to the cultured black pearl industry. The survival of turbot eggs and the rearing capacities of larvae stemmed from artificial fertilization practices using frozen-thawed spermatozoa were evaluated. Furthermore, the viability of sperm samples stored during a 9 month period in liquid nitrogen was assessed. No significant difference in the fertilization rate, hatching rate, survival and wet weight of 10-day old larvae were observed using fresh or frozen-thawed spermatozoa. A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm.
